A search engine will see the hotspot URL as it's in the HTML code for the page.
There is a lot of folklore about SEO based on speculation and on assuming search engines have more intelligence than they probably have.
Who knows if search engines take more notice of hyperlinks from individual images than to hotspots applied over an image?
WebPlus hotspots are applied to a 1x1 pixel transparent GIF image stretched to a larger area. Hopefully search engines will not spot that!
